Thioctic acid — better known as lipoic acid — is a small sulfur-bearing molecule with an unusual property: it is soluble in both water and fat. Most antioxidants can only work in one compartment. Lipoic acid moves through both, which has earned it the description of a universal antioxidant, and it can regenerate other antioxidants — vitamins C and E, glutathione — after they have been spent.
Its primary job, though, is enzymatic. Lipoic acid is the covalently bound cofactor of the multi-enzyme complexes that catalyze the oxidative decarboxylation of pyruvate and alpha-ketoglutarate to acetyl coenzyme A and succinyl-CoA. Those two reactions are the entrance to the citric acid cycle and a key step within it. Without lipoic acid, carbohydrate cannot be committed to aerobic energy production at all.
The body synthesizes small amounts in the mitochondria, where it is made and used on the spot. Supplemental lipoic acid is free rather than protein-bound, and it is this free form that circulates and acts as an antioxidant.
